Abstract Drug combination therapy is generally more effective than monotherapy in the field of cancer treatment. However, screening for effective synergistic combinations from a wide range of drug combinations is particularly important given the increase in the number of available drug classes and potential drug-drug interactions. Existing methods for predicting the synergistic effects of drug combinations primarily focus on extracting structural features of drug molecules and cell lines, but neglect the interaction mechanisms between cell lines and drug combinations. Consequently, there is a deficiency in comprehensive understanding of the synergistic effects of drug combinations. To address this issue, we propose a drug combination synergy prediction model based on multi-source feature interaction learning, named MFSynDCP, aiming to predict the synergistic effects of anti-tumor drug combinations. This model includes a graph aggregation module with an adaptive attention mechanism for learning drug interactions and a multi-source feature interaction learning controller for managing information transfer between different data sources, accommodating both drug and cell line features. Comparative studies with benchmark datasets demonstrate MFSynDCP's superiority over existing methods. Additionally, its adaptive attention mechanism graph aggregation module identifies drug chemical substructures crucial to the synergy mechanism. Overall, MFSynDCP is a robust tool for predicting synergistic drug combinations. The source code is available from GitHub at https://github.com/kkioplkg/MFSynDCP .

Chemotherapy-induced mucositis represents a severe adverse outcome of cancer treatment, significantly curtailing the efficacy of these treatments and, in some cases, resulting in fatal consequences. Despite identifying intestinal epithelial cell damage as a key factor in chemotherapy-induced mucositis, the paucity of effective treatments for such damage is evident. In our study, we discovered that Eubacterium coprostanoligenes promotes mucin secretion by goblet cells, thereby fortifying the integrity of the intestinal mucus barrier. This enhanced barrier function serves to resist microbial invasion and subsequently reduces the inflammatory response. Importantly, this effect remains unobtrusive to the anti-tumor efficacy of chemotherapy drugs. Mechanistically, E. copr up-regulates the expression of AUF1, leading to the stabilization of Muc2 mRNA and an increase in mucin synthesis in goblet cells. An especially significant finding is that E. copr activates the AhR pathway, thereby promoting the expression of AUF1. In summary, our results strongly indicate that E. copr enhances the intestinal mucus barrier, effectively alleviating chemotherapy-induced intestinal mucositis by activating the AhR/AUF1 pathway, consequently enhancing Muc2 mRNA stability.

Abstract Arising from the extreme/saddle point in electronic bands, Van Hove singularity (VHS) manifests divergent density of states (DOS) and induces various new states of matter such as unconventional superconductivity. VHS is believed to exist in one and two dimensions, but rarely found in three dimension (3D). Here, we report the discovery of 3D VHS in a topological magnet EuCd2As2 by magneto-infrared spectroscopy. External magnetic fields effectively control the exchange interaction in EuCd2As2, and shift 3D Weyl bands continuously, leading to the modification of Fermi velocity and energy dispersion. Above the critical field, the 3D VHS forms and is evidenced by the abrupt emergence of inter-band transitions, which can be quantitatively described by the minimal model of Weyl semimetals. Three additional optical transitions are further predicted theoretically and verified in magneto-near-infrared spectra. Our results pave the way to exploring VHS in 3D systems and uncovering the coordination between electronic correlation and the topological phase.

Objective: To evaluate the application value of combining spectral imaging and metal artifact reduction (MAR) in head and neck CTA after the embolization of intracranial aneurysms. Methods: We collected 37 patients who experienced embolization of intracranial aneurysms then received spectral imaging of head and neck CTA. Monochromatic images with energy ranging from 70～140 keV, 120 kVp-like mixed energic images, 70～140 keV MAR images, and 120 kVp-like MAR images were generated. The region of interest was placed on the area near the coil and with the most serious metal artifact. CT attenuation and standard deviation were measured, and artifact index (AI) and signal-noise ratio (SNR) were calculated. Two radiologists independently subjectively evaluated the metal artifact and the display of surrounding vessels using Likert 5 scales. The subjective scores and objective parameters between MAR and non-MAR images were compared. The Wilcoxon ranking test, paired sample t test, and independent sample t test were utilized to compare parameters between the groups. Results: MAR images had significantly lower AI than did non-MAR images for all eight monochromatic energies. When energies ranged from 80~110 keV, SNR was higher for MAR images than for non-MAR images, and the difference was statistically significant. With same energies, MAR images had higher artifact and vessel display scores than did non-MAR images. For non-MAR images, the different coil diameters did not make a statistical difference in AI and vessel display scores. For MAR images, a larger coil diameter (>8.79 mm) led to higher AI and lower vessel display scores than did normal diameters (≤8.79 mm). Conclusion: The combination of spectral imaging and MAR could effectively reduce the metal artifact of implants for the embolization of intracranial aneurysms and improve the surrounding vessel display. Moreover, the metal artifact reduction effect was more significant for the coils with smaller diameters.

Abstract Background Focusing on key indicators of drought resistance is highly important for quickly mining candidate genes related to drought resistance in cotton. Results In the present study, drought resistance was identified in drought resistance-related RIL populations during the flowering and boll stages, and multiple traits were evaluated; these traits included three key indicators: plant height (PH), single boll weight (SBW) and transpiration rate (Tr). Based on these three key indicators, three groups of extreme mixing pools were constructed for BSA-seq. Based on the mapping interval of each trait, a total of 6.27 Mb QTL intervals were selected on chromosomes A13 (3.2 Mb), A10 (2.45 Mb) and A07 (0.62 Mb) as the focus of this study. Based on the annotation information and qRT‒PCR analysis, three key genes that may be involved in the drought stress response of cotton were screened: GhF6'H1, Gh3AT1 and GhPER55. qRT‒PCR analysis of parental and extreme germplasm materials revealed that the expression of these genes changed significantly under drought stress. Cotton VIGS experiments verified the important impact of key genes on cotton drought resistance. Conclusions This study focused on the key indicators of drought resistance, laying the foundation for the rapid mining of drought-resistant candidate genes in cotton and providing genetic resources for directed molecular breeding of drought resistance in cotton.

Abstract To evaluate the corrective effect of posterior hemivertebra resection and short-segment fusion surgery on pediatric patients and to assess the impact of short-segment fixation surgery on vertebral development during follow-up, a retrospective analysis was performed on 28 pediatric patients who underwent posterior hemivertebra resection surgery. The corrective effect was evaluated by comparing indicators such as segmental scoliosis Cobb angle, upper and lower compensatory curves and trunk balance at different time points. Meanwhile, the vertebral and spinal canal diameters of instrumented vertebrae and adjacent noninstrumented vertebrae were measured and compared to assess vertebral and spinal canal development. The correction rate of segmental scoliosis was 72.2%. The estimated mean vertebral volume of the instrumented vertebra was slightly lower than that of the unfused segment at the final follow-up, but the difference was not statistically significant. The growth rate of the spinal canal during follow-up was much smaller than that of the vertebral body. In summary, internal fixation at a young age shows no significant inhibitory effects on spinal development within the fusion segment. Posterior hemivertebra resection and short-segment fusion surgery are safe and effective.

Abstract Background This study aimed to explore the association between ideal cardiovascular health metrics (ICVHM) and arthritis (AR), as well as the interactions of various indicators in ICVHM on AR in US adults. Methods We involved 17,041 participants who were interviewed by NHANES from 2011 to 2018. AR included osteoarthritis or degenerative arthritis (OA), rheumatoid arthritis (RA), and psoriatic arthritis and other arthritis (Other AR). Logistic regression was applied to analyze the association between AR and ICVHM. Mixed graphical model (MGM) was used to explore the interaction between variables in ICVHM. Results Higher ICVHM scores had a protective effect on AR. Compared to “≤1” score, the ORs of AR in participants with 2, 3, 4, and ≥5 were 0.586, 0.472, 0.259, and 0.130, respectively. Similar results were also found in different types of AR. ICVHM has a maximum area under the curve value of 0.765 and the interaction between blood pressure and total cholesterol was 0.43. Conclusions ICVHM correlates significantly with AR and is better at identifying AR than individual indicators. ICVHM can be better improved by controlling the indicators with stronger interactions. Our findings provide guidance for promoting health factors, which have important implications for identification and prevention of AR.

ObjectiveProbiotics are beneficial to the intestinal barrier, but few studies have investigated probiotics from giant pandas. This study aims to explore the preventive effects of giant panda-derived Clostridium butyricum on dextran sodium sulfate (DSS)-induced colitis in mice.MethodsClostridium butyricum was administered to mice 14 days before administering DSS treatment to induce enteritis.ResultsClostridium butyricum B14 could more effectively prevent colitis in mice than C. butyricum B13. C. butyricum B14 protected the mouse colon by decreasing the histology index and serum interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-α) levels, which improved intestinal inflammation-related symptoms. In addition, the treatment led to the regulation of the expression of Tifa, Igkv12-89, and Nr1d1, which in turn inhibited immune pathways. The expression of Muc4, Lama3, Cldn4, Cldn3, Ocln, Zo1, Zo2, and Snai is related the intestinal mucosal barrier. 16S sequencing shows that the C. butyricum B14 significantly increased the abundance of certain intestinal probiotics. Overall, C. butyricum B14 exerted a preventive effect on colitis in mice by inhibiting immune responses, enhancing the intestinal barrier and increasing the abundance of probiotic species. Thus, C. butyricum B14 administration helps regulate the balance of the intestinal microecology. It can suppress immune pathways and enhance barrier-protective proteins.

Abstract Background Current research suggests that there is an association between housing tenure and the health status of migrant populations, but the potential mediators of this association remain to be explored. We aimed to examine the effect of housing tenure on migrant populations’ health in China and how this effect is mediated by health service utilization and social integration. Methods Data from the 2017 China Migrants Dynamic Survey of 47,459 participants was used. Logistic regression models were used to explore the effects of housing tenure, health service utilization, and social integration on the health status of migrant populations. Mediated effects models were used to explore the association among them. This study used the bootstrap method and KHB method to test the mediating effect of health service utilization and social integration. Results Compared to private renters, owners with mortgages (OR: 0.828, 95% CI: 0.765–0.896) were significantly associated with a higher risk of poor health. Compared with private renters, outright owners were associated with a lower risk of poor health (OR: 1.016, 95% CI: 0.935, 1.104), but the association was not statistically significant (p > 0.05). Abstract. Background:. Osteopenia has been well documented in adolescent idiopathic scoliosis (AIS). Bone marrow stem cells (BMSCs) are a crucial regulator of bone homeostasis. Our previous study revealed a decreased osteogenic ability of BMSCs in AIS-related osteopenia, but the underlying mechanism of this phenomenon remains unclear. Methods:. A total of 22 AIS patients and 18 age-matched controls were recruited for this study. Anthropometry and bone mass were measured in all participants. Bone marrow blood was collected for BMSC isolation and culture. Osteogenic and adipogenic induction were performed to observe the differences in the differentiation of BMSCs between the AIS-related osteopenia group and the control group. Furthermore, a total RNA was extracted from isolated BMSCs to perform RNA sequencing and subsequent analysis. Results:. A lower osteogenic capacity and increased adipogenic capacity of BMSCs in AIS-related osteopenia were revealed. Differences in mRNA expression levels between the AIS-related osteopenia group and the control group were identified, including differences in the expression of LRRC17, DCLK1, PCDH7, TSPAN5, NHSL2, and CPT1B. Kyoto Encyclopedia of Genes and Genomes enrichment analyses revealed several biological processes involved in the regulation of autophagy and mitophagy. The Western blotting results of autophagy markers in BMSCs suggested impaired autophagic activity in BMSCs in the AIS-related osteopenia group. Conclusion:. Our study revealed that BMSCs from AIS-related osteopenia patients have lower autophagic activity, which may be related to the lower osteogenic capacity and higher adipogenic capacity of BMSCs and consequently lead to the lower bone mass in AIS patients.

ABSTRACT: Objectives: The incidence of Helicobacter pylori (HP) is 25–50% in developed countries and 80% in developing countries, including 56.2% in China. However, antibiotic resistance of HP is a threat to HP control. The purpose of this study was to comprehensively evaluate primary drug resistance of HP in China. Methods: The full text of reports of the primary antibiotic resistance prevalence of HP was obtained from multiple databases (PubMed, Web of Science, Evimed, Cochrane Library, and China National Knowledge Internet). Review Manager 5.2 was adopted for meta-analysis, sensitivity analysis, and bias analysis. The Newcastle-Ottawa Scale was used to assess the article quality. Results: In total, 38804 HP samples from 22 trials were extracted. The results suggested that the overall prevalence of amoxicillin, clarithromycin, metronidazole, and levofloxacin resistance among HP in adults was as follows: mean difference (MD) = 1.35%, 95% confidence interval (CI) [1.03%, 1.68%]; MD = 23.76%, 95% CI [20.23%, 27.3%]; MD = 69.32%, 95% CI [64.85%, 73.8%]; and MD = 29.45%, 95% CI [4.90, 176.96], respectively. From the results of sensitivity and publication bias, we find that these results are robust and had little publication bias. Conclusion: Our research showed that in China, the prevalence of HP resistance to primary antibiotics warrants attention, especially with regard to metronidazole, levofloxacin, and clarithromycin.

Abstract Purpose The study aimed to retrospectively identify the prognostic factors of surgically treated primary tongue squamous cell carcinoma (TSCC) cases and assess the benefits of surgical neck lymph node dissection (LND) in early‐stage cancer. Methods Patients with primary TSCC with pT1‐2N0‐1M0 stage without distant metastasis who were treated with surgery during 2014–2016 at Xiangya Hospital, Central South University were included. Univariate and multivariate Cox models were constructed to explore prognostic factors of overall survival (OS), disease‐free survival (DFS), and local recurrence‐free survival (LRFS). Sub‐group analysis was used to assess the effect of adjuvant therapy and the prognostic value of LND for the early‐stage patients. Results In total, 440 patients met the inclusion criteria. During the follow‐up period, the 5‐year OS, DFS, were 84.4% and 70.0%, respectively. Univariate analysis showed that TNM stage, lymphovascular invasion (LVI), and/or perineural invasion (PNI), pathological differentiation, etc. were significant predictors of OS and DFS. Multivariate analysis showed that TNM stage and the degree of pathological differentiation were independent prognostic factors for all outcomes. Besides, the number of cervical LND could independently predict both DFS and LRFS while LVI/PNI were associated with DFS. And high‐quality neck LND (≥30) significantly improved DFS and LRFS for patients of pT1cN0M0 or stage I as compared to those without LND. Conclusions TNM stage and pathological differentiation were crucial prognostic factors for postoperative patients with TSCC. Notably, high‐quality cervical LND was beneficial for the improvement of DFS and LRFS for patients of pT1cN0M0 or stage I.

The escalation in carbon dioxide concentration has precipitated global climate warming, accentuating ecological and environmental concerns. Notably, China stands as the world’s largest carbon emitter, with the Taihu Lake basin emerging as a carbon-intensive region within the country. This paper undertakes a comprehensive analysis spanning 2005 to 2020, calculating the economic contribution coefficient of carbon emissions and the ecological carrying coefficient of carbon absorption in the Taihu Lake basin. The study includes a delineation of carbon balance zones and an exploration of the geographical and spatial influences of both ecosystem and economic factors. The overarching trend in carbon emissions within the Taihu Lake Basin initially exhibited rapid growth, followed by a fluctuating decline, with the pivotal year being 2012, recording the apex of emissions at 575.8293 million tons. Concurrently, total carbon absorption demonstrated a fluctuating growth trajectory, ascending from 82.3503 million tons in 2005 to 85.6488 million tons in 2020. The carbon emission intensity in the basin manifested a pattern of high concentration in the northeast and low concentration in the southwest, while the carbon absorption intensity displayed the inverse pattern. The carbon balance across the Taihu Lake Basin revealed a spatial incongruity, characterized by a suboptimal pattern in the northeast and a favorable pattern in the southwest. Zhejiang Province emerged as an ecological stronghold within the basin, acting as the primary carbon sink functional area. Urban built-up areas and forested regions emerged as principal influencers of carbon balance in the Taihu Lake basin. Urban construction land, population density, and arable land area were identified as primary contributors to carbon emissions, whereas per capita GDP, forests, grasslands, and water bodies were identified as main contributors to carbon absorption in the watershed.

IntroductionMicrobiome within plant tissues is pivotal for co-evolution with host plants. This microbiome can colonize the plant, with potential transmission via seeds between parents and offspring, affecting seedling growth and host plant adaptability to the environment.MethodsWe employed 16S rRNA gene amplicon analysis to investigate the vertical distribution of core microbiome in cotton seeds across ecological niches [rhizosphere, root, stem, leaf, seed and seed-P (parental seed)] of the three cotton genotypes.ResultsThe findings demonstrated a significant decrease in microbiome diversity and network complexity from roots, stems, and leaves to seeds. The microenvironment exerted a more substantial influence on the microbiome structure of cotton than the genotypes. The core endophytic microorganisms in cotton seeds comprised 29 amplicon sequence variants (ASVs) affiliated with Acidimicrobiia, Alphaproteobacteria, Bacilli, Bacteroidia, Clostridia, Gammaproteobacteria, and unclassified_Proteobacteria. These vertically transmitted taxa are widely distributed in cotton plants. Through 16S rRNA gene-based function prediction analysis of the cotton microbiome, we preliminarily understood that there are potential differences in metabolic capabilities and phenotypic traits among microbiomes in different microhabitats.DiscussionIn conclusion, this study demonstrated the crucial role of the microenvironment in influencing the cotton microbiome and offered insights into the structures and functions of the cotton seed microbiome, facilitating future crop yield enhancement through core seed microbiome regulation.

Objective: We aim to explore the association between the Composite Dietary Antioxidant Index (CDAI), physical activity, and cognitive impairment. Methods: 2395 participants from the 2011–2014 National Health and Nutrition Examination Survey (NHANES) were included in this study. The association of CDAI and physical activity with cognitive impairment was analyzed using binary logistic regression. A stratified analysis was employed for a sensitivity analysis of the logistic regression, examining the interaction between CDAI and each stratified variable. Results: Both low scores on CDAI and physical inactivity are associated with higher prevalence of cognitive impairment. Compared to the fourth quartile of CDAI and active group, the prevalence of cognitive impairment is statistically increased in other sub-groups. Conclusion: There is an interaction between CDAI and physical activity. The elderly population with low scores on CDAI especially combined with physical inactivity should be focused, so as to reduce the adverse effects of cognitive impairment.

To address this challenge and optimize Immune check inhibitor treatment, understanding the underlying molecular intricacies involved is crucial. The emergence of CRISPR-Cas9 technology has empowered researchers to precisely investigate gene function and has introduced transformative shifts in identifying key genes for various physiological and pathological processes. CRISPR screenings, particularly in vivo CRISPR screenings, have become invaluable tools in deciphering molecular networks and signaling pathways governing suppressive immune checkpoint molecules. In this review, we provide a comprehensive overview of in vivo CRISPR screenings in cancer immunotherapy, exploring how this cutting-edge technology has unraveled potential novel therapeutic targets and combination strategies. We delve into the latest findings and advancements, shedding light on immune checkpoint regulation and offering exciting prospects for the development of innovative and effective treatments for cancer patients.

Diabetic wounds (DWs) pose a major challenge for the public health system owing to their high incidence, complex pathogenesis, and long recovery time; thus, there is an urgent need to develop innovative therapies to accelerate the healing process of diabetic wounds. As natural nanovesicles, extracellular vesicles (EVs) are rich in sources with low immunogenicity and abundant nutritive molecules and exert potent therapeutic effects on diabetic wound healing. To avoid the rapid removal of EVs, a suitable delivery system is required for their controlled release. Owing to the advantages of high porosity, good biocompatibility, and adjustable physical and chemical properties of hydrogels, EV biopotentiated hydrogels can aid in achieving precise and favorable therapy against diabetic wounds. This review highlights the different design strategies, therapeutic effects, and mechanisms of EV biopotentiated hydrogels. We also discussed the future challenges and opportunities of using EV biopotentiated hydrogels for diabetic wound healing.

Speech emotion recognition (SER) is an active research area in affective computing. Recognizing emotions from speech signals helps to assess human behaviour, which has promising applications in the area of human-computer interaction. The performance of deep learning-based SER methods relies heavily on feature learning. In this paper, we propose SCAR-NET, an improved convolutional neural network, to extract emotional features from speech signals and implement classification. This work includes two main parts: First, we extract spectral, temporal, and spectral-temporal correlation features through three parallel paths; and then split-convolve-aggregate residual blocks are designed for multi-branch deep feature learning. The features are refined by global average pooling (GAP) and pass through a softmax classifier to generate predictions for different emotions. We also conduct a series of experiments to evaluate the robustness and effectiveness of SCAR-NET which can achieve 96.45%, 83.13%, and 89.93% accuracy on the speech emotion datasets EMO-DB, SAVEE, and RAVDESS. These results show the outperformance of SCAR-NET.

Abstract. Background:. Congenital scoliosis (CS) is a complex spinal malformation of unknown etiology with abnormal bone metabolism. Fibroblast growth factor 23 (FGF23), secreted by osteoblasts and osteocytes, can inhibit bone formation and mineralization. This research aims to investigate the relationship between CS and FGF23. Methods:. We collected peripheral blood from two pairs of identical twins for methylation sequencing of the target region. FGF23 mRNA levels in the peripheral blood of CS patients and age-matched controls were measured. Receiver operator characteristic (ROC) curve analyses were conducted to evaluate the specificity and sensitivity of FGF23. The expression levels of FGF23 and its downstream factors fibroblast growth factor receptor 3 (FGFr3)/tissue non-specific alkaline phosphatase (TNAP)/osteopontin (OPN) in primary osteoblasts from CS patients (CS-Ob) and controls (CT-Ob) were detected. In addition, the osteogenic abilities of FGF23-knockdown or FGF23-overexpressing Ob were examined. Results:. DNA methylation of the FGF23 gene in CS patients was decreased compared to that of their identical twins, accompanied by increased mRNA levels. CS patients had increased peripheral blood FGF23 mRNA levels and decreased computed tomography (CT) values compared with controls. The FGF23 mRNA levels were negatively correlated with the CT value of the spine, and ROCs of FGF23 mRNA levels showed high sensitivity and specificity for CS. Additionally, significantly increased levels of FGF23, FGFr3, OPN, impaired osteogenic mineralization and lower TNAP levels were observed in CS-Ob. Moreover, FGF23 overexpression in CT-Ob increased FGFr3 and OPN levels and decreased TNAP levels, while FGF23 knockdown induced downregulation of FGFr3 and OPN but upregulation of TNAP in CS-Ob. Mineralization of CS-Ob was rescued after FGF23 knockdown. Conclusions:. Our results suggested increased peripheral blood FGF23 levels, decreased bone mineral density in CS patients, and a good predictive ability of CS by peripheral blood FGF23 levels. FGF23 may contribute to osteopenia in CS patients through FGFr3/TNAP/OPN pathway.

Water scarcity is one of the significant constraints on sustainable agricultural development in arid and semi-arid regions. The challenges faced in forage production are even more severe than those encountered with general crops. The industry still struggles to achieve water-efficient, high-yield quality forage in water-scarce pastoral areas. This study focuses on alfalfa, a high-quality forage crop, employing a combination of “subsurface drip irrigation (SDI) + alternate partial root-zone irrigation (APRI)” and establishing three water supply gradients (full irrigation, 75% deficit, 50% deficit), in comparison with the widely used subsurface drip irrigation, to study the effects of two irrigation methods and three moisture gradients on alfalfa. The aim is to provide some theoretical basis and data support for achieving water-saving and high-yield quality forage in water-scarce pastoral areas. The main findings are as follows: First, compared with SDI, the two-year alternate dry and wet environment provided by alternate partial root-zone drip irrigation (ARDI) significantly increased the specific root length, specific surface area, and root length density of alfalfa at 20~40 cm depth, increasing by 33.3~76.8%, 6.4~32.97%, and 15.2~93.9%, respectively, compared to SDI. Under ARDI irrigation, the alfalfa root system has a greater contact area with the soil, which lays a solid foundation for the water and nutrient supply needed for the accumulation of its above-ground biomass. Secondly, over the two-year production process, the plant height of alfalfa under ARDI treatment was 12~14.5% higher than that under SDI, the total fresh forage yield was 43.5~64% higher, and the total dry forage yield was 23.2~33.8% higher than SDI. Under ARDI, the 75% water deficit treatment could still maintain the plant height and stem thickness of alfalfa compared to full irrigation with SDI and increased the dry forage yield by 6.6% without significantly reducing the quality, significantly enhancing the productive performance of alfalfa. Moreover, during the two years of production and utilization, the nutritional quality of alfalfa under the ARDI irrigation mode did not significantly decrease compared to SDI, maintaining the stable nutritional quality of alfalfa over multiple years of production. Lastly, thanks to the improved root system and increased yield of alfalfa under ARDI irrigation, and based on this, its water evapotranspiration did not significantly increase compared to SDI; the annual average Alfalfa Water Productivity Index (AWPI) and Alfalfa Water Productivity of Crop (AWPC) under ARDI irrigation increased by 28.8% and 37.2%, respectively, improving the water use efficiency of alfalfa production. In summary, in the production of alfalfa in water-scarce pastoral areas, ARDI and its water deficit treatment have more potential for water-saving than SDI as a water-saving irrigation strategy.

Systemic vasculitides are a rare and complex group of diseases that can affect multiple organ systems. Clinically, presentation may be vague and non-specific and as such, diagnosis and subsequent management are challenging. These entities are typically classified by the size of vessel involved, including large-vessel vasculitis (giant cell arteritis, Takayasu’s arteritis, and clinically isolated aortitis), medium-vessel vasculitis (including polyarteritis nodosa and Kawasaki disease), and small-vessel vasculitis (granulomatosis with polyangiitis and eosinophilic granulomatosis with polyangiitis). There are also other systemic vasculitides that do not fit in to these categories, such as Behcet’s disease, Cogan syndrome, and IgG4-related disease. Advances in medical imaging modalities have revolutionized the approach to diagnosis of these diseases. Specifically, color Doppler ultrasound, computed tomography and angiography, magnetic resonance imaging, positron emission tomography, or invasive catheterization as indicated have become fundamental in the work up of any patient with suspected systemic or localized vasculitis. This review presents the key diagnostic imaging modalities and their clinical utility in the evaluation of systemic vasculitis.

Background Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is the central enzyme of glycolysis and plays important regulatory roles in plant growth and development and responses to adverse stress conditions. However, studies on the characteristics and functions of cotton GAPDH family genes are still lacking. Methods In this study, genome-wide identification of the cotton GAPDH gene family was performed, and the phylogeny, gene structures, promoter progenitors and expression profiles of upland cotton GAPDH gene family members were explored by bioinformatics analysis to highlight potential functions. The functions of GhGAPDH9 in response to drought stress were initially validated based on RNA-seq, qRT‒PCR, VIGS techniques and overexpression laying a foundation for further studies on the functions of GAPDH genes. Results This study is the first systematic analysis of the cotton GAPDH gene family, which contains a total of 84 GAPDH genes, among which upland cotton contains 27 members. Quantitative, phylogenetic and covariance analyses of the genes revealed that the GAPDH gene family has been conserved during the evolution of cotton. Promoter analysis revealed that most cis-acting elements were related to MeJA and ABA. Based on the identified promoter cis-acting elements and RNA-seq data, it was hypothesized that Gh_GAPDH9, Gh_GAPDH11, Gh_GAPDH19 and Gh_GAPDH21 are involved in the response of cotton to abiotic stress. The expression levels of the Gh_GAPDH9 gene in two drought-resistant and two drought-sensitive materials were analyzed by qRT‒PCR and found to be high early in the treatment period in the drought-resistant material. The silencing of Gh_GAPDH9 based on virus-induced gene silencing (VIGS) technology resulted in significant leaf wilting or whole-plant dieback in silenced plants after drought stress compared to the control. The content of—malondialdehyde (MDA) in cotton leaves was significantly increased, and the content of proline (Pro) and chlorophyll (Chl) was reduced. In addition, the leaf wilting and dryness of transgenic lines under drought stress were lower than those of wild-type Arabidopsis, indicating that Gh_GAPDH9 is a positive regulator of drought resistance. In conclusion, our results demonstrate that GAPDH genes play an important role in the response of cotton to abiotic stresses and provide preliminary validation of the function of the Gh_GAPDH9 gene under drought stress. These findings provide an important theoretical basis for further studies on the function of the Gh_GAPDH9 gene and the molecular mechanism of the drought response in cotton.

Abstract Background The advancements in unmanned aerial vehicle (UAV) technology have recently emerged as an effective, cost-efficient, and versatile solution for monitoring crop growth with high spatial and temporal precision. This monitoring is usually achieved through the computation of vegetation indices (VIs) from agricultural lands. The VIs are based on the incoming radiance to the camera, which is affected when there is a change in the scene illumination. Such a change will cause a change in the VIs and subsequent measures, e.g., the VI-based chlorophyll-content estimation. In an ideal situation, the results from VIs should be free from the impact of scene illumination and should reflect the true state of the crop’s condition. In this paper, we evaluate the performance of various VIs computed on images taken under sunny, overcast and partially cloudy days. To improve the invariance to the scene illumination, we furthermore evaluated the use of the empirical line method (ELM), which calibrates the drone images using reference panels, and the multi-scale Retinex algorithm, which performs an online calibration based on color constancy. For the assessment, we used the VIs to predict leaf chlorophyll content, which we then compared to field measurements. Results The results show that the ELM worked well when the imaging conditions during the flight were stable but its performance degraded under variable illumination on a partially cloudy day. For leaf chlorophyll content estimation, The $$r^2$$ r 2 of the multivariant linear model built by VIs were 0.6 and 0.56 for sunny and overcast illumination conditions, respectively. The performance of the ELM-corrected model maintained stability and increased repeatability compared to non-corrected data. The Retinex algorithm effectively dealt with the variable illumination, outperforming the other methods in the estimation of chlorophyll content. The $$r^2$$ r 2 of the multivariable linear model based on illumination-corrected consistent VIs was 0.61 under the variable illumination condition. Conclusions Our work indicated the significance of illumination correction in improving the performance of VIs and VI-based estimation of chlorophyll content, particularly in the presence of fluctuating illumination conditions.

Abstract Phosphatidylinositol 3-kinase alpha (PI3Kα) inhibitors are currently evaluated for the therapy of esophageal squamous cell carcinoma (ESCC). It is of great importance to identify potential biomarkers to predict or monitor the efficacy of PI3Kα inhibitors in an aim to improve the clinical responsive rate in ESCC. Here, ESCC PDXs with CCND1 amplification were found to be more sensitive to CYH33, a novel PI3Kα-selective inhibitor currently in clinical trials for the treatment of advanced solid tumors including ESCC. Elevated level of cyclin D1, p21 and Rb was found in CYH33-sensitive ESCC cells compared to those in resistant cells. CYH33 significantly arrested sensitive cells but not resistant cells at G1 phase, which was associated with accumulation of p21 and suppression of Rb phosphorylation by CDK4/6 and CDK2. Hypo-phosphorylation of Rb attenuated the transcriptional activation of SKP2 by E2F1, which in turn hindered SKP2-mediated degradation of p21 and reinforced accumulation of p21. Moreover, CDK4/6 inhibitors sensitized resistant ESCC cells and PDXs to CYH33. These findings provided mechanistic rationale to evaluate PI3Kα inhibitors in ESCC patients harboring amplified CCND1 and the combined regimen with CDK4/6 inhibitors in ESCC with proficient Rb.

As an excellent chicken breed found in a high-altitude zone of northern China, Lindian chickens are characterized by good egg and meat production, strong adaptability, cold tolerance, rough feeding resistance, excellent egg quality, and delicious meat quality. To facilitate the exploitation of the unique qualities of the Lindian chicken, the varying patterns and correlations of various body size and carcass traits of 3–22-week-old Lindian chickens were analyzed in this study. The optimal growth model of these traits was determined by growth curve fitting analysis. The results showed that most traits of Lindian chickens increased steadily with increasing age, and most of them increased rapidly before 10 weeks of age. In addition, the inflection point age of each trait was predicted to be between 4 and 10 weeks. Furthermore, this study revealed that body size traits were closely related to carcass traits in Lindian chickens. In summary, Lindian chickens are in a rapid growth stage before the age of 10 weeks, and better slaughter performance can be achieved through good feeding management during this stage. The reproductive traits and muscles are the main developmental focus after the age of 19 weeks, so it is important to adequately meet their energy requirements for subsequent good breeding performance.

In this paper, a distributed cooperative control problem for a class of high-order multi-input and multi-output (MIMO) nonlinear multiagent systems (MASs) in the presence of uncertain nonlinearities and external disturbances is addressed. A coupled design is developed to collaboratively approximate unknown nonlinearities and compounded disturbances by combining neural networks (NNs) with high-order disturbance observers (HODOs). To further simplify the controller structure, relationships among the Laplace matrix, adjacency matrix and consensus tracking errors are analyzed based on undirected communication graphs. Then, a distributed adaptive NN anti-disturbance control protocol is proposed for high-order MIMO nonlinear MASs based on the outputs of NNs and HODOs, where dynamic surface control (DSC) is introduced to eliminate the “computational explosion” problem of the conventional backstepping method. The semiglobally uniformly ultimate boundedness of closed-loop system signals is proven through Lyapunov theory. Finally, simulations of a quadrotor UAV formation are performed to demonstrate the effectiveness of the proposed control scheme.

Advances in transcriptomic technologies have deepened our understanding of the cellular gene expression programs of multicellular organisms and provided a theoretical basis for disease diagnosis and therapy. However, both bulk and single-cell RNA sequencing approaches lose the spatial context of cells within the tissue microenvironment, and the development of spatial transcriptomics has made overall bias-free access to both transcriptional information and spatial information possible. Here, we elaborate development of spatial transcriptomic technologies to help researchers select the best-suited technology for their goals and integrate the vast amounts of data to facilitate data accessibility and availability. Then, we marshal various computational approaches to analyze spatial transcriptomic data for various purposes and describe the spatial multimodal omics and its potential for application in tumor tissue. Finally, we provide a detailed discussion and outlook of the spatial transcriptomic technologies, data resources and analysis approaches to guide current and future research on spatial transcriptomics.

BackgroundDespite numerous observational studies linking adiposity, diabetes, and lifestyle factors with gliomas, the causal associations between them remain uncertain.MethodsThis study aimed to use two-sample Mendelian randomization (MR) analysis to investigate whether these associations are causal. Specifically, independent genetic variants in body mass index (BMI), waist circumference (WC), type 2 diabetes (T2D), smoking, alcohol, and coffee consumption were extracted from the published genome-wide association studies (GWASs) with genome-wide significance. The corresponding summary-level data for gliomas were available from a GWAS of 1,856 cases and 4,955 controls of European descent from the GliomaScan consortium. Additionally, glioma pathogenesis-related protein 1 data were used for validation, and Radial MR analysis was conducted to examine the potential outlier single-nucleotide polymorphisms (SNPs).ResultsOne standard deviation (SD) increase in BMI had an odds ratio (OR) of 1.392 (95% confidence interval (CI), 0.935–2.071) for gliomas, while one SD increase in WC had an OR of 0.967 (95% CI, 0.547–1.710). For T2D, a one-unit increase in log-transformed OR had an OR of 0.923 (95% CI, 0.754–1.129). The prevalence of smoking initiation had an OR of 1.703 (95% CI, 0.871–3.326) for gliomas, while the prevalence of alcohol intake frequency had an OR of 0.806 (95% CI, 0.361–1.083), and the prevalence of coffee intake had an OR of 0.268 (95% CI, 0.033–2.140) for gliomas.ConclusionThis study provides evidence that adiposity, T2D, smoking, alcohol drinking, and coffee intake do not play causal roles in the development of gliomas. The findings highlight the importance of reconsidering causal relationships in epidemiological research to better understand the risk factors and prevention strategies for gliomas.

Umbilical cord-derived mesenchymal stem cells (UC-MSC) are promising candidates for wound healing. However, the low amplification efficiency of MSC in vitro and their low survival rates after transplantation have limited their medical application. In this study, we fabricated a micronized amniotic membrane (mAM) as a microcarrier to amplify MSC in vitro and used mAM and MSC (mAM-MSC) complexes to repair burn wounds. Results showed that MSC could live and proliferate on mAM in a 3D culture system, exhibiting higher cell activity than in 2D culture. Transcriptome sequencing of MSC showed that the expression of growth factor-related, angiogenesis-related, and wound healing-related genes was significantly upregulated in mAM-MSC compared to traditional 2D-cultured MSC, which was verified via RT-qPCR. Gene ontology (GO) analysis of differentially expressed genes (DEGs) showed significant enrichment of terms related to cell proliferation, angiogenesis, cytokine activity, and wound healing in mAM-MSC. In a burn wound model of C57BL/6J mice, topical application of mAM-MSC significantly accelerated wound healing compared to MSC injection alone and was accompanied by longer survival of MSC and greater neovascularization in the wound.

Abstract The high-affinity K+ transporter (HAK) family plays a vital role in K+ uptake and transport as well as in salt and drought stress responses. In the present study, we identified 22 HAK genes in each Medicago truncatula and Medicago sativa genome. Phylogenetic analysis suggested that these HAK proteins could be divided into four clades, and the members of the same subgroup share similar gene structure and conserved motifs. Many cis-acting elements related with defense and stress were found in their promoter region. In addition, gene expression profiles analyzed with genechip and transcriptome data showed that these HAK genes exhibited distinct expression pattern in different tissues, and in response to salt and drought treatments. Furthermore, co-expression analysis showed that 6 homologous HAK hub gene pairs involved in direct network interactions. RT-qPCR verified that the expression level of six HAK gene pairs was induced by NaCl and mannitol treatment to different extents. In particular, MtHK2/7/12 from M. truncatula and MsHAK2/6/7 from M. sativa were highly induced. The expression level of MsHAK1/2/11 determined by RT-qPCR showed significantly positive correlation with transcriptome data. In conclusion, our study shows that HAK genes play a key role in response to various abiotic stresses in Medicago, and the highly inducible candidate HAK genes could be used for further functional studies and molecular breeding in Medicago.

Abstract Background The biological phenotype of tumours evolves during neoadjuvant chemotherapy (NAC). Accurate prediction of pathological complete response (pCR) to NAC in the early-stage or posttreatment can optimize treatment strategies or improve the breast-conserving rate. This study aimed to develop and validate an autosegmentation-based serial ultrasonography assessment system (SUAS) that incorporated serial ultrasonographic features throughout the NAC of breast cancer to predict pCR. Methods A total of 801 patients with biopsy-proven breast cancer were retrospectively enrolled from three institutions and were split into a training cohort (242 patients), an internal validation cohort (197 patients), and two external test cohorts (212 and 150 patients). Three imaging signatures were constructed from the serial ultrasonographic features before (pretreatment signature), during the first–second cycle of (early-stage treatment signature), and after (posttreatment signature) NAC based on autosegmentation by U-net. The SUAS was constructed by subsequently integrating the pre, early-stage, and posttreatment signatures, and the incremental performance was analysed. Results The SUAS yielded a favourable performance in predicting pCR, with areas under the receiver operating characteristic curve (AUCs) of 0.927 [95% confidence interval (CI) 0.891–0.963] and 0.914 (95% CI 0.853–0.976), compared with those of the clinicopathological prediction model [0.734 (95% CI 0.665–0.804) and 0.610 (95% CI 0.504–0.716)], and radiologist interpretation [0.632 (95% CI 0.570–0.693) and 0.724 (95% CI 0.644–0.804)] in the external test cohorts. Furthermore, similar results were also observed in the early-stage treatment of NAC [AUC 0.874 (0.793–0.955)–0.897 (0.851–0.943) in the external test cohorts]. Conclusions We demonstrate that autosegmentation-based SAUS integrating serial ultrasonographic features throughout NAC can predict pCR with favourable performance, which can facilitate individualized treatment strategies.

Abstract Background Psoriasis has been linked to cardiovascular disease (CVD), including coronary artery disease (CAD), myocardial infarction (MI), and heart failure (HF). However, available studies regarding this relationship have shown inconsistent results. Therefore, in this report, we performed a comprehensive review of the literature to assess the effects of psoriasis on risk of these CVDs. Methods A search of literature until 24 December 2021 was done in PubMed, the Cochrane Library, Web of Science, Google Scholar, and Embase. Within European and East Asian populations, meta-analyses of observational studies assessing correlations between psoriasis and various CVD risk factors were conducted. Mendelian randomization (MR) was then employed to assess the causative impact of genetic pre-disposition for psoriasis on these CVD risk factors. Results The results of the meta-analyses indicated that, in both the European and East Asian populations, psoriasis was significantly linked to an elevated risk in the incidence of CAD (RR = 1.51, 95% confidence interval (CI): 1.04–2.18, p = 0.028 and RR = 1.91, 95% CI: 1.62–2.25, p

Abstract Active Peltier cooling enables Peltier heat transfer in addition to the traditional Fourier thermal conductance, which is useful in some special applications, such as the microthermostats. From the material wise, however, the study on the active Peltier cooling materials is rare. We carried out a high-throughput workflow to screen out 5 room-temperature active Peltier cooling materials, GaSbLi2, HgPbCa2, SnTiRu2, GeYbLi2, and GeTiFe2, from 2958 Heusler materials. All the five materials are semimetals or very narrow band gap systems with high electrical conductivity. Some of these materials have relatively large Seebeck coefficients due to the band asymmetry. Their effective thermal conductivity κ eff s, which are the summation of active Peltier thermal conductivity and passive thermal conductivity, are all greater than Cu at the room temperature and ΔT = 1 K. The present work gives a possible way to search active cooling Peltier materials for the applications of precise temperature control.

Abstract Background Serine carboxypeptidase-like protein (SCPL) plays an important role in response to stress in plant. However, our knowledge of the function of the SCPL gene family is limited. Results In this study, a comprehensive and systematic analysis of SCPL gene family was conducted to explore the phylogeny and evolution of the SCPL gene in Gossypium hirsutum. The phenotype and molecular mechanism of silencing of the Gh_SCPL42 under Verticillium wilt stress was also studied. Our results showed that 96 SCPL genes were observed in genome of G. hirsutum, which distributed on 25 chromosomes and most of them were located in the nucleus. The phylogenetic tree analysis showed that members of SCPL gene family can be divided into three subgroups in G. hirsutum, which are relatively conservative in evolution. SCPL gene has a wide range of tissue expression types in G. hirsutum. Promoter analysis showed that the most cis-acting elements related to MeJA and ABA were contained. Through RNA-seq combined with genotyping, it was found that 11 GhSCPL genes not only had significant expression changes during Verticillium wilt stress but also had differential SNPs in the upstream, downstream, exonic or intronic regions. The expression of these 11 genes in the resistant (Zhongzhimian 2) and susceptible (Junmian 1) materials was further analyzed by qRT-PCR, it was found that 6 genes showed significant expression differences in the two materials. Among them, Gh_SCPL42 has the most obvious expression change. Furthermore, virus-induced gene silencing (VIGS) showed necrosis and yellowing of leaves and significantly higher disease severity index (DSI) and disease severity rate (DSR) values in VIGS plants than in control silenced Gh_SCPL42 plants. Moreover, the expression levels of genes related to the SA and JA pathways were significantly downregulated. These results show that Gh_SCPL42 might improve resistance to Verticillium wilt through the SA and JA pathways in G. hirsutum. Conclusion In conclusion, our findings indicated that Gh_SCPL42 gene plays an important role in resistance to Verticillium wilt in cotton. It was provided an important theoretical basis for further research on the function of SCPL gene family and the molecular mechanism of resistance to Verticillium wilt in cotton.

The survival of Escherichia coli (E. coli) bacteria, the most common faecal indicator bacteria (FIB), may be significantly affected by cyanobacteria present during a harmful algal bloom (HAB). Therefore, the effect of Microcystis on the survival of FIB E.coli and coliforms was investigated. Microcosms containing two species of Microcystis (M. aeruginosa and M. smithii) were established and then inoculated with four reference strains of E. coli (ATCC 25922, 8739, 51813, and 11775) to explore the cyanobacteria-bacteria dynamics at a laboratory setting. Monitoring over several days showed normal growth of Microcystis, with or without the presence of E. coli. However, Microcystis was shown to dramatically decrease the survival of E. coli over time. Analysis of microcystin production by Microcystis was found to correlate with loss of E. coli, suggesting a toxic effect of microcystins on E. coli bacteria. This phenomenon was also demonstrated for a natural consortium of E. coli and coliform bacteria by inoculating with contaminated lake water. The results indicate that the use of E. coli as FIB may be greatly compromised in the presence of Microcystis spp. such as during a HAB when associated toxins are produced.

Synthesis of methylsilsesquioxane aerogels by ambient pressure drying instead of supercritical drying has recently emerged as a major trend, but the issues of low mechanical strength and unstable performance still need to be resolved. This work reveals the microscopic formation mechanisms of gel skeleton based on the kinetic characteristics of methyltrimethoxysilane (MTMS) precursor hydrolysis and the associated sol-gel reactions. The effects of oxalic acid concentration (cOA) and hydrolysis time of MTMS solution (th) on the gelation time, morphologies, microstructures, chemical structure, and compression properties of the as-synthesized methylsilsesquioxane aerogels are investigated. The optimal cOA and th are 38.4 mmol/L and 120 min, respectively, endowing the methylsilsesquioxane aerogels with a compression strength of 0.170 MPa and a maximum compression strain of 61.2%. Precise control of the hydrolysis conditions ensures the formation of branched particle-to-particle networks, which is crucial for maximizing the compression properties of methylsilsesquioxane aerogels synthesized under industry-relevant conditions.

Fibrosis is a common healing process that occurs during stress and injury in cardiovascular diseases. The evolution of fibrosis is associated with cardiovascular disease states and causes adverse effects. Fibroblast activation is responsible for the formation and progression of fibrosis. The incipient detection of activated fibroblasts is important for patient management and prognosis. Fibroblast activation protein (FAP), a membrane-bound serine protease, is almost specifically expressed in activated fibroblasts. The development of targeted FAP-inhibitor (FAPI) positron emission tomography (PET) imaging enabled the visualisation of FAP, that is, incipient fibrosis. Recently, research on FAPI PET imaging in cardiovascular diseases increased and is highly sought. Hence, we comprehensively reviewed the application of FAPI PET imaging in cardiovascular diseases based on the state-of-the-art published research. These studies provided some insights into the value of FAPI PET imaging in the early detection of cardiovascular fibrosis, risk stratification, response evaluation, and prediction of the evolution of left ventricular function. Future studies should be conducted with larger populations and multicentre patterns, especially for response evaluation and outcome prediction.

Knowledge graphs (KGs) have gained prominence for representing real-world facts, with queries of KGs being crucial for their application. Aggregate queries, as one of the most important parts of KG queries (e.g., “ What is the average price of cars produced in Germany?”), can provide users with valuable statistical insights. An efficient solution for KG aggregate queries is approximate aggregate queries with semantic-aware sampling (AQS). This balances the query time and result accuracy by estimating an approximate aggregate result based on random samples collected from a KG, ensuring that the relative error of the approximate aggregate result is bounded by a predefined error. However, AQS is tailored for simple aggregate queries and exhibits varying performance for complex aggregate queries. This is because a complex aggregate query usually consists of multiple simple aggregate queries, and each sub-query influences the overall processing time and result quality. Setting a large error bound for each sub-query yields quick results but with a lower quality, while aiming for high-quality results demands a smaller predefined error bound for each sub-query, leading to a longer processing time. Hence, devising efficient and effective methods for executing complex aggregate queries has emerged as a significant research challenge within contemporary KG querying. To tackle this challenge, we first introduced an execution cost model tailored for original AQS (i.e., supporting simple queries) and founded on Taylor’s theorem. This model aids in identifying the initial parameters that play a pivotal role in the efficiency and efficacy of AQS. Subsequently, we conducted an in-depth exploration of the intrinsic relationship of the error bounds between a complex aggregate query and its constituent simple queries (i.e., sub-queries), and then we formalized an execution cost model for complex aggregate queries, given the accuracy constraints on the error bounds of all sub-queries. Harnessing the multi-objective optimization genetic algorithm, we refined the error bounds of all sub-queries with moderate values, to achieve a balance of query time and result accuracy for the complex aggregate query. An extensive experimental study on real-world datasets demonstrated our solution’s superiority in effectiveness and efficiency.

Abstract Background An Andersson lesion (AL) is a fatigue fracture occurring across three columns in ankylosing spondylitis (AS), resulting in spinal pseudarthrosis (SP) formation, most commonly in the thoracolumbar segment. However, there is still great controversy and few reports on the best surgical method for the treatment of AS combined with thoracolumbar AL. The purpose of this study was to investigate the efficacy of posterior closed osteotomy, debridement and fusion through the fracture line for the treatment of this disease. Methods The clinical data of 13 patients (male 8, female 5, mean age 50.6 years) with AS combined with thoracolumbar AL treated with posterior closed osteotomy, debridement and fusion through the fracture line were retrospectively analysed. The following parameters of the full-length lateral spine radiographs were measured preoperatively and at the last follow-up: cervical 7 tilt (C7T), global kyphosis (GK), thoracic kyphosis (TK), thoracolumbar kyphosis (TLK), local kyphosis (LK), angle of the fusion levels (AFL), lumbar lordosis (LL), pelvic incidence (PI), pelvic tilt (PT), sacral slope (SS) and sagittal vertical axis (SVA). The visual analog scale (VAS), Oswestry disability index (ODI) and Scoliosis Research Society-22 (SRS-22) scores were recorded preoperatively and at the last follow-up. Results The mean operation time was 345 min, the mean blood loss was 673 mL, and the mean follow-up time was 21.9 months. Abstract Background The recurrence of bile duct stones is a long‐term outcome for patients undergoing laparoscopic common bile duct exploration (LCBDE) that is worthy of attention. This study aimed to investigate long‐term risk factors for stones recurrence after LCBDE and develop a nomogram for predicting the risk. Methods The clinical data on consecutive patients with bile duct stones undergoing LCBDE at Shanghai Tenth People's Hospital between January 2014 and February 2019 with a follow‐up period longer than 2 years were reviewed. Independent risk factors of stones recurrence identified by the Cox regression model were used to develop a nomogram in predicting stones recurrence after LCBDE. Results Eight hundred and twenty‐two patients were eventually included in this study. Of these patients, 42 (5.11%) developed stones recurrence. The cumulative incidences of stones recurrence at 1, 3, and 5 years after LCBDE were 1.34%, 4.36%, and 7.14%, respectively. Independent risk factors of stones recurrence were identified to be age (HR = 1.04, 95% CI = 1.02‐1.07), T‐tube drainage (HR = 3.28, 95% CI = 1.23‐8.72), fatty liver (HR = 2.69, 95% CI = 1.39‐5.20), urinary calculus (HR = 4.68, 95% CI = 2.29‐9.56), post‐cholecystectomy (HR = 5.21, 95% CI = 2.39‐11.33), and post‐ERCP + EST (HR = 2.87, 95% CI = 1.18‐6.96). By these factors, a developed nomogram showed a C‐index of 0.770 to predict stones recurrence. Conclusions The nomogram, based on identified risk factors, showed good accuracy for predicting stones recurrence, which is valuable to guide these patients’ follow‐up and prevention.

In order to explore the degradation of imidacloprid, acetamiprid and triazophos by dielectric barrier discharge low-temperature plasma, the aqueous solution was established to study the effects of the discharge voltage, discharge time, initial pesticide concentration and pH of solution on degradation. The degradation kinetics model was also analyzed and the degradation path was proposed based on the degradation products identification. The results showed that the low-temperature plasma technology could effectively degrade the three pesticide residues in the aqueous solution and triazophos was much more labile to low-temperature plasma treatment than imidacloprid and acetamiprid. Under the conditions of this study, the increase of discharge voltage and discharge time, and the decrease of initial pesticides concentration benefitted the increase of degradation rate. Alkaline conditions were conducive to the degradation of imidacloprid and acetamiprid, and acidic conditions benefitted the degradation of triazophos. The maximum degradations rates were 62.5% for imidacloprid (pH9.0), 42.4% for acetamiprid (pH9.0) and 94.5% for triazophos (pH3.0) after the treatment at 13.6 kV for 5 min at the initial concentration of 1.9 mg/L. The degradation kinetics of all pesticides were fitted to the first-order kinetics model well (R2≥0.90). Seven and five degradation products of imidacloprid and acetamiprid were identified respectively. The formation of imidacloprid and acetamiprid degradation products mainly experienced the breaking of C-H, C-N bonds and the oxidation and substitution of hydroxyl radicals.

Abstract In the context of big data, the exploration of the application effect of machine learning in intelligent encryption for real-time image text digital information aims to improve the privacy information security of people. Aiming at the problem of digital information leakage of real-time image text, the convolutional neural network is introduced and improved by adding a preprocessing module to form AlexNet, to encrypt the digital information of real-time image text. Besides, to take into account both the security effect and the real-time performance of the system, the image text is encrypted by the chaotic sequence generated by a one-dimensional chaotic system called Logistic-Sine and a multi-dimensional chaotic system named Lorenz. In this way, a real-time image text encryption model is constructed by combining the chaotic function and AlexNet. Finally, a simulation experiment is performed to analyze the performance of this model. The comparative analysis indicates that the recognition accuracy of feature extraction of image text by the intelligent encryption model reaches 94.37%, which is at least 3.05% higher than that of other neural network models by scholars in related fields. In the security analysis of image text encryption, the information entropy of pixel values at (0, 0) of the proposed model is close to the ideal value 8. Meanwhile, the value of the number of pixels change rate is generally more than 99.50%, and the value of the unified average changing intensity is generally more than 33.50%. This demonstrates that the model has good security in resisting attacks. Therefore, the constructed model can provide good security guarantee under the premise of ensuring the recognition accuracy, which can provide experimental basis for improving the security performance of real-time image text data in the future.

Esophageal cancer is one of the most lethal cancers worldwide because of its rapid progression and poor prognosis. Esophageal squamous cell carcinoma (ESCC) and esophageal adenocarcinoma (EAC) are two major subtypes of esophageal cancer. ESCC predominantly affects African and Asian populations, which is closely related to chronic smoking and alcohol consumption. EAC typically arises in Barrett's esophagus with a predilection for Western countries. While surgical operation and chemoradiotherapy have been applied to combat this deadly cancer, molecularly targeted therapy is still at the early stages. With the development of large-scale next-generation sequencing, various genomic alterations in ESCC and EAC have been revealed and their potential roles in the initiation and progression of esophageal cancer have been studied. Potential therapeutic targets have been identified and novel approaches have been developed to combat esophageal cancer. In this review, we comprehensively analyze the genomic alterations in EAC and ESCC and summarize the potential role of the genetic alterations in the development of esophageal cancer. Progresses in the therapeutics based on the different tissue types and molecular signatures have also been reviewed and discussed.

BackgroundNuclear protein of the testis (NUT) carcinoma is a rare subset of poorly differentiated, highly aggressive malignancy defined by NUTM1 gene rearrangements. Only three NUT cases of probable ovarian origin have been reported.Case presentationWe report a case of NUT carcinoma in a 53-year-old female who presented with extensive abdominopelvic lesions and bilateral ovarian masses suggestive of advanced ovarian cancer. This patient was admitted to our hospital due to abdominal pain and distension for over two months. Imaging examinations suggested a possible malignancy of bilateral adnexal origin. This patient first underwent diagnostic laparoscopy. After receiving neoadjuvant chemotherapy, she underwent cytoreductive surgery. Surgical pathology showed infiltration of monotonous round tumor cells with no apparent differentiation characteristics. Immunohistochemistry (IHC) revealed nuclear expression of the NUT protein. And MXI1::NUTM1 fusion was identified by next-generation sequencing (NGS). Herein, we introduce an unusual NUT carcinoma and describe the clinical, imaging, and pathological features. In addition, we briefly reviewed the published literature and discussed the possibility of primary gynecological NUT carcinoma.ConclusionsIdentifying a NUT carcinoma arising from the abdominopelvic cavity is essential, and we underscore the need for NUT testing in undifferentiated malignant neoplasms that appear in this clinical setting. Although it is unclear from which origin this tumor arose, proper classification is essential for treatment planning.

Smallpox was eradicated in less than 200 years after Edward Jenner’s practice of cowpox variolation in 1796. The forty-three years of us living free of smallpox, beginning in 1979, never truly separated us from poxviruses. The recent outbreak of monkeypox in May 2022 might well warn us of the necessity of keeping up both the scientific research and public awareness of poxviruses. One of them in particular, the vaccinia virus (VACV), has been extensively studied as a vector given its broad host range, extraordinary thermal stability, and exceptional immunogenicity. Unceasing fundamental biological research on VACV provides us with a better understanding of its genetic elements, involvement in cellular signaling pathways, and modulation of host immune responses. This enables the rational design of safer and more efficacious next-generation vectors. To address the new technological advancement within the past decade in VACV research, this review covers the studies of viral immunomodulatory genes, modifications in commonly used vectors, novel mechanisms for rapid generation and purification of recombinant virus, and several other innovative approaches to studying its biology.
